砷广泛存在于自然环境中,是一种具有毒性的类金属[1]。环境中砷的来源包括自然来源和人为来源。自然环境中,砷存在于地壳、火成岩和沉积岩中,主要以黄铁矿、雄黄、雌黄砷等硫化矿物的形式存在[2]。而人为活动,如化学肥料、杀虫剂的使用,以及采矿活动,都会促进砷在地下水和农田中的积累[3]。砷因为其剧毒性,被美国毒物与疾病登记署列为一类危险物质,危险程度高于镉、铅、汞等有毒金属[4]。长期处于高砷的环境,会对人体健康造成严重伤害[5]。
稻田砷污染引起的生态风险和食品安全问题,日渐引起人们的广泛关注。砷可以通过水稻秸秆和稻米经食物链进入动物和人体。水稻是南亚和东南亚的主要粮食产物,食用稻米和饮用水是摄入砷的主要途径[6]。相比其他谷物,稻米及其副产品的食用可能导致摄入过量的无机砷,对人体健康造成损害。由于水稻生长期间的间歇性淹水排水和水稻生理生态特性,不论是在砷污染与否的土壤中,水稻谷粒中积累的砷比其他谷物都要高,大约是其他谷物砷含量的10倍[7]。砷在水稻土中主要以无机砷——砷(Ⅲ)、砷(Ⅴ)的形式存在,而水稻谷粒中的无机砷主要为砷(Ⅲ),甲基化砷主要为二甲基砷(DMA(Ⅴ))[8]。无机砷对水稻本身毒性不高,对水稻生长影响不大,但是对人体却有剧毒性,特别是砷(Ⅲ);而甲基砷对人体毒性较低,但却对水稻有很大的毒害作用,影响水稻的正常生长[9]。
微生物修复作为新兴的修复技术,具有效果好、经济、不会产生二次污染的优势,已被证明是有效的修复方法,受到了国内外研究者的关注[10]。因此,本文将阐明微生物在稻田环境中影响砷循环的过程,总结微生物在稻田砷生物化学循环过程中扮演的角色,了解微生物在水稻生态系统中影响砷转化的机理,为进一步研发稻田砷污染修复技术提供参考。
1 微生物介导下砷的环境化学行为砷在土壤中通过水稻根部的硅酸盐和磷酸盐转运通道进入水稻,一部分砷被储存在根部的液泡中,另一部分砷则经木质部导管向地上部运输,进入水稻茎叶部分,再通过韧皮部的介导进入水稻籽粒中。水稻根部吸收的砷不断向水稻各部分组织中运输,其砷含量表现为:根 > 茎叶 > 谷壳 > 糙米,在整个生长期间,水稻各组织部位砷含量逐渐增加[11]。减少水稻根部对砷的吸收,是控制砷进入水稻籽粒的关键[12]。土壤中砷形态的变化影响着砷的移动性、毒性和生物有效性,对水稻根部吸收砷有很大的影响。
砷(Ⅲ)的活性、毒性和迁移能力均强于砷(Ⅴ),且有着更强的附着能力和移动性,因而更易被植物吸收[13]。因此,砷(Ⅴ)酸盐的还原通常导致水稻土壤中砷的活化,并提高水稻对砷的利用率。相反,亚砷(Ⅲ)酸盐的氧化则利于砷的固定。微生物介导的砷代谢过程在植物–土壤–微生物系统的砷循环中发挥着重要作用,包括砷(Ⅲ)氧化、砷(Ⅴ)还原和砷(Ⅲ)甲基化。微生物不仅可以改变稻田砷的形态,进而影响水稻对砷的利用程度,还可以通过影响铁、硫的循环过程以间接影响水稻生态系统中砷的形成和迁移[14]。
砷抗性微生物可以通过自身的生物转化途径来影响砷的生物化学行为,主要是因为其体内存在多种基因编码、转化酶和转运体,可以在体内对砷进行氧化、还原、甲基化和挥发[15]。根据微生物对砷氧化还原的作用不同,可以将这类微生物分为砷还原微生物、砷氧化微生物和砷甲基化微生物(表 1)。
对砷具有还原功能的微生物分为两种:一种是异养型砷还原微生物,可以在无氧环境下将砷酸盐作为电子供体,以各种有机物,如氢、葡萄糖甲酸盐等及少数芳香族化合物作为电子受体,通过呼吸还原作用将砷还原,并能从中获得能量,供自身的细胞生长[11];另一种是耐砷微生物,当环境砷浓度过高时,它们可以将砷还原排出细胞外作为自身的解毒机制来保障自身的生存,不能为自身的生长提供能量[20]。
异养型砷还原微生物对砷(Ⅴ)的还原机制,也被称之为呼吸还原机制。在呼吸还原过程中,主要由具有催化作用的呼吸还原蛋白(ARR)参与[41]。有学者对C. arsenatis[42]、Bacillus selenitireducens[43]、Shewanella strain ANA-3[44]微生物进行提纯分析,均发现了结构为异构二聚体的呼吸还原蛋白酶,它主要由两部分构成:一个钼蛋白大亚基(ArrA)和以[4Fe-4S]聚簇为中心的小亚基(ArrB)[45]。这种呼吸还原酶主要存在于细胞外膜的外周胞质中,可以与周围结合态的砷如铁氧化物、铁锰氧化物等增加接触的机会,因此能够还原吸附在固体介质上的砷,将结合态的砷还原成迁移性高的游离态砷。在这个过程中,砷(Ⅴ)与ArrA进行结合还原成砷(Ⅲ)并释放出电子,ArrB再将电子传递给c型细胞色素(cytc2)[46]。Malasarn等[44]对革兰氏阴性菌ANA-3中编码ArrA的保守基因arrA进行了克隆测序,并进一步证明了arrA为砷酸盐还原酶基因,是异化砷酸盐微生物在呼吸作用过程中还原砷(Ⅴ)的可靠标志,但其是否直接参与砷的释放过程,目前仍不清楚[47]。
大部分耐砷微生物对砷的解毒过程为:砷(Ⅴ)通过磷酸盐转运体(pit和pst)进入细胞体内,将砷(Ⅴ)还原成砷(Ⅲ),通过排出蛋白将砷(Ⅲ)排出细胞外。在这个过程中主要需要砷还原酶ArsC和外排泵的参与。Silver等[48]通过对从大肠杆菌分离出的ArsC酶进行分析,发现在砷还原过程中还需要还原性谷胱甘肽(GSH)或硫醇蛋白–谷氧还蛋白(Grx)作为电子供体。砷(Ⅴ)进入细胞内与位于ArsC N-末端的半胱氨酸残基结合,被还原性GSH或Grx提供的电子还原为砷(Ⅲ),然后利用ArsA蛋白水解释放ATP提供能量,使亚砷酸盐通过ArsB形成的亚砷酸盐转运体排出细胞外,从而形成一个挤压系统,将砷排出细胞外,以此减轻砷对自身的毒害[45]。arsC基因广泛存在于好氧、厌氧微生物中,而arrA主要存在于厌氧微生物中。在淹水条件下,水稻根际环境中的arsC基因在丰度和多样性上均高于arrA基因,表明在稻田生态系统中,arsC基因在影响砷转化的过程中扮演着重要角色[49]。
1.2 微生物对砷的氧化行为化能自养型砷氧化微生物能够在好氧和厌氧条件下,将砷(Ⅲ)氧化成毒性较低和移动性较差的砷(Ⅴ)。它们可以分别以O2或NO3-作为电子受体,将砷(Ⅲ)作为电子供体氧化砷(Ⅲ),并利用这个过程中产生的能量进行CO2的固定和细胞的生长[50]。近年来,对化能自养型微生物的砷氧化酶及其相关编码基因的研究取得了一定的成果。Zhang等[19]从砷污染稻田中分离出新的化能自养型砷氧化微生物Paracoccus niistensis SY,通过基因分析发现,aioA基因负责氧化酶亚基A的编码,aioA基因的缺失使该菌株的砷(Ⅲ)氧化能力消失,说明aioA基因在表达砷氧化酶中占据重要位置。Aio操纵子结构基因由负责编码大亚基AioA和小亚基AioB亚砷酸氧化酶的基因aioA/aioB、cytc2和钼掺杂素生物合成基因(chlE)组成。Wang等[24]研究表明,AioE可能作为一种新型的砷(Ⅲ)氧化电子转运体与NADH的产生有关,对砷的氧化还原有重要的影响,其编码基因aioE位于aioBA氧化酶基因和砷抗性基因arsR1-arsC1-arsC2- acr3-1附近,砷(Ⅲ)氧化过程中产生的电子可能通过AioE传递给AioA/B再传递给cytc2。Zargar等[51]从Alkalilimnicola ehrlichii strain MLHE-1中分离得到了一种新的氧化酶ArxA,由基因arxA编码而成,仅在厌氧条件下表达;对16S rRNA基因和aroA基因的系统发育进行分析,结果表明,arsA与砷还原基因arrA有很大的同源性,而且有报道[52]表明,微生物MLHE-1的arxA在体外可逆亚砷酸盐氧化和砷酸盐还原中发挥作用。
异养型砷氧化微生物同样能够氧化砷(Ⅲ),但与化能自养型砷氧化微生物不同的是,它并不能从氧化砷的过程中获得能量,而是需要有机物质作为提供能量和细胞物质的来源,如Alcaligenes sp. strain RS - 19、Thermus 3As、Thermus HR13[38]。异养型砷氧化微生物通过利用外周胞质上的砷氧化酶,将细胞周围的砷(Ⅲ)氧化,降低细胞周围砷的毒性,被理解为是对砷中毒的一种解毒的表现。近年来,异养型砷氧化微生物的氧化酶和基因的有关研究取得了一定的进展[53]。分析A. faecalis NCIB8687和Hydrogenophaga NT - 14发现,这种外周胞质酶是由两个亚单位构成的:AoxB和AoxA。AoxB作为一个大亚单位是由[3Fe-4S]聚簇中心和两个钼辅酶蛋白共同构成的,而AoxA是由Rieske-[2Fe-2S]聚簇的一个小亚单位[31]。研究表明,这种酶属DMSO还原酶家族的一员,由操纵子aoxA和aoxB控制[28]。
除砷氧化菌和砷还原菌外,自然界中还存在一类同时兼备砷氧化和砷还原的微生物。如Zhang等[54]首次从砷污染的土壤中分离到一株革兰氏阴性菌Pseudomonas sp. HN-2,它们在好氧条件下能在3 h内将92% 的砷(Ⅲ)氧化成砷(Ⅴ);而在厌氧条件下,其可以将砷(Ⅴ)还原成砷(Ⅲ)。Thermus HR13可以有效地将砷(Ⅲ)氧化成砷(Ⅴ),大约是非生物速率的100倍;在氧气缺乏的情况下,它还能利用乳酸和砷(Ⅴ)呼吸,还原砷(Ⅴ)。Thermus HR13同样具有砷氧化和砷还原的功能[55]。
1.3 微生物对砷的甲基化行为稻田体系中砷甲基化是一种微生物介导的过程,发生在植物吸收之前的土壤中。一些微生物能够通过在体内合成砷甲基转移酶,包括真菌、细菌、真核藻类和古生菌进行砷甲基化作用,可以将砷依次添加一个甲基基团,生成毒性较低的有机砷,依次为单、二、三甲基砷化合物,最终产物为具有挥发性的三甲基胂(TMAO)。不同形态的砷,毒性不同,毒性大小顺序为:二甲基亚胂酸(DMA(Ⅲ)) > 一甲基亚胂酸(MMA(Ⅲ)) > 砷(Ⅲ) > 砷(Ⅴ) > 二甲基胂(DMA(Ⅴ)) > 一甲基胂酸(MMA(Ⅴ)) > 三甲基胂(TMAO)[56]。微生物对砷进行甲基化的产物主要为DMA(Ⅴ)和TMAO,毒性均比无机砷低,后者且具有挥发性。砷被挥发至空气中,在空气中稀释,浓度大大降低,因此挥发性砷的形成能够有效地降低土壤和水体中砷的含量。有研究表明,每年大约有2.1×107 kg的砷由于微生物的作用从土壤中挥发到大气层[57]。arsM基因是最早从微生物鉴定分离出来的控制砷甲基化的基因[56]。水稻籽粒中甲基化砷占籽粒中总砷的10% ~ 90%,水稻根际土壤和非根际土中甲基化砷的浓度与土壤中微生物arsM基因丰度呈正相关,此结果以及arsM基因水平转移的证据表明,arsM基因在水稻土壤中分布广泛,这也是为什么大米远比其他谷物含有更高浓度砷甲基化物的原因[58]。除arsM基因可以对砷进行甲基化外,在最近的研究当中,Verma等[59]分别在真菌和丛枝菌根真菌(AMF)中,分离鉴定出可以将砷甲基化为MMA、DMA和最终挥发性TMAO的新的砷酸盐甲基转移酶基因WaarsM和RiMT-11。因此,微生物对砷的甲基化作用被认为是最为理想的砷修复的生物途径,微生物驱动的砷转化可能降低水稻–土壤–水系统中砷的生物有效性。
1.4 微生物介导铁循环对砷形态的影响在自然条件下,土壤中存在许多的铁氧化物,包括铁的氢氧化物和氧化物两大类铁矿物[60]。铁矿物对砷具有强的亲和力,能够强烈地吸附土壤中游离的砷,从而降低砷的迁移能力及毒性。Bennett等[61]研究发现,相较铁(Ⅱ),铁(Ⅲ)对砷具有更强的吸附能力,土壤溶液中的铁(Ⅱ)与砷(Ⅲ)浓度呈极显著正相关关系,铁矿物的还原溶解可以影响砷的生物活性。水稻根际环境中铁(Ⅲ)/砷的比率要远高于周围土壤中铁(Ⅲ)/砷的比率[62],这主要是因为水稻根际的通气组织能够向根部进行径向泌氧,向根际环境释放氧气,可以把周围的铁(Ⅱ)氧化成铁(Ⅲ),生成对砷(Ⅲ)和砷(Ⅴ)具有较强吸附能力的铁(Ⅲ),在水稻根际形成一层以铁氢氧化物为主的铁膜[63]。铁膜通过与砷酸盐在表面发生螯合作用,将砷(Ⅲ)和砷(Ⅴ)吸附在铁矿物表面,对砷进入水稻起到一个屏障的作用,降低水稻对砷的吸收,相反,当铁膜中铁(Ⅲ)被还原,会使固定的砷活化释放出来,增加砷的生物可利用度。因此,铁(Ⅲ)水平越高,水稻土–水系统中生物可利用砷的浓度越低。
在自然情况下,铁矿物的砷解吸是一个缓慢的过程。微生物是影响铁矿物吸收释放砷的重要驱动力[64]。土壤中存在铁的氧化细菌(FeOB)能够将铁(Ⅱ)氧化成铁(Ⅲ),在水稻土壤中,铁(Ⅱ)的氧化主要依靠3种微生物,包括光合铁氧化菌、微嗜氧铁氧化菌和厌氧硝酸还原菌[65]。在厌氧环境中,铁(Ⅱ)主要依靠光养性铁氧化菌(如R. ferrooxidans SW2、R. palustris strain TIE-1等)和硝态氮还原铁氧化菌(如Thiobacillus denitrificans、T. denitrificans、Acidovorax sp. strain 2AN等)进行铁(Ⅱ)氧化,形成具有吸附性及可与砷共沉淀的铁(Ⅲ)氢氧化物[66-67]。光养性铁氧化菌在氧化过程中需要碳酸盐作为电子受体还原碳源,铁(Ⅱ)作为电子供体,在光能的条件下进行铁氧化。大部分硝态氮铁氧化菌氧化铁(Ⅱ)的过程需要在有机共底物环境下进行,如醋酸,伴随着NO3-的反硝化,NO3-还原成NO、NO2和N2,某一程度上可以将铁氧化物视为微生物反硝化作用的副产品[62]。在土壤含氧量较低时,铁(Ⅱ)的氧化主要依靠微嗜氧铁氧化菌(如Gallionella spp.,Mariprofundus spp. strains等)进行,其可以以氧作为电子受体,铁(Ⅱ)作为电子供体,通过氧化铁(Ⅱ)来获得生长所需的能量;而在高氧浓度下,均质非生物铁(Ⅱ)氧化同时发生,形成含铁(Ⅲ)矿物产物,由于此类铁(水)氧化物与砷(Ⅴ)和砷(Ⅲ)都有很强的亲和力,它们显著地降低了砷在水稻土–水系统中的迁移率和生物可利用度,以及随后在水稻组织中吸收和积累的砷[68]。
相反地,也会存在铁的还原菌(FeRB),可以将铁(Ⅲ)还原成铁(Ⅱ)[41, 69]。古菌域和细菌域中几乎均存在着FeRB。FeRB是铁异化还原的主要动力。在代谢过程中,FeRB释放电子,利用氧化铁作为电子受体,将铁(Ⅲ)还原成铁(Ⅱ)。铁膜中铁(Ⅲ)的还原,会使砷释放出来,提高砷的迁移率和生物利用度。不同的FeRB对铁(Ⅲ)进行异化还原具有不同的机制:①直接接触:FeRB可以在细胞外形成具导电性和电子传递能力的胞外聚合物,与铁(Ⅲ)氧化物直接接触,还原铁(Ⅲ);②铁螯合剂作用:FeRB通过分泌铁(Ⅲ)螯合物,加速铁(Ⅲ)氧化物的溶解,进而促进铁(Ⅲ)还原;③电子穿梭体作用:FeRB利用氧化还原性物质,如腐殖质,与不溶性铁(Ⅲ)进行电子传递,还原铁(Ⅲ);④菌毛作用:又称纳米导线,连接FeRB与电极间的桥梁,可进行电子远距离传递,使细菌对铁(Ⅲ)氧化物产生趋向性,实现铁(Ⅲ)的还原[70](图 1)。
砷的行为受硫(S)的非生物或生物氧化还原作用的影响。硫的氧化还原作用可以使土壤和矿物中固持的砷释放,也可以使土壤中砷固定。硫化物具有很强的还原性,当土壤环境中的氧化还原电势从–120 mV降低到–180 mV时,SO42-可以还原成S2-[71],对砷的溶解度和迁移率的变化具有很大的影响。硫酸盐还原菌(SRB)可以促进SO42-还原成S2-,铁(Ⅲ)和砷(Ⅴ)可以被S2-还原,生成铁(Ⅱ)和砷(Ⅲ),从铁砷固相中释放出来,提高了砷的生物有效性[72]。但当环境中砷(Ⅲ)/硫摩尔比≥0.22、存在足够的硫酸盐时,S2-可以与砷(Ⅲ)发生耦合反应生成硫化砷物质——H3AsO2S2、H3AsOS3、H3AsO3S和As2S3,形成更稳定的硫化物矿物,重新固定砷,或与铁(Ⅱ)结合,生成硫化铁(即FeS或FeS 2)作为二次沉淀物,之后,砷(Ⅲ)则可与新生成的二次沉淀物通过吸附或共沉淀被再次固定,形成二次铁矿物FeAsS,有效地降低砷的释放和有效性[73]。SRB可以改变铁矿物的形态,降低铁矿物的结晶度,从而增加二次铁矿物对砷的吸附能力[74](图 1)。
2 砷污染的微生物修复微生物修复是利用微生物对重金属吸附、沉淀、氧化和挥发作用来降低重金属浓度和毒性的一项技术[10]。微生物修复在水体污染方面有很广泛的应用。例如在各种污水处理系统中,细菌对污水的降解起着至关重要的作用[76]。微生物对重金属的转化能力还可被利用在农作物安全生产中。微生物在水稻生态系统中,可以通过驱动砷的转化,来降低水稻对砷的植物利用度,缓解水稻受砷污染影响。在稻田中接种微藻不仅可以促进水稻的生长,还可以调节磷酸盐的转运,有效地降低水稻谷粒中砷的含量[10, 77]。Debnath和Bhadury[78]从稻田中分离出5种固氮蓝藻,可以有效地将砷(Ⅲ)转化成砷(Ⅴ),并可以将大量的砷酸盐固定在藻细胞中,减少水稻对砷的吸收,降低砷对水稻的毒性。此外,集胞藻、念珠藻和微囊藻等常见蓝藻具有甲基转移酶基因,可将无机砷转化为有机砷MMA(Ⅲ) 挥发[79]。刘云霞等[80]就接种和不接种菌根真菌(Glomus mosseae)对早稻中砷积累的影响研究表明,接种菌根真菌能够明显提高早稻地上部和地下部对磷的利用,磷从地下部向地上部转运能力的提高有效抑制了砷从地下部到地上部的传输,降低地上部和地下部中砷的积累,这与菌根真菌降低土壤溶液中砷(Ⅲ)进而降低水稻砷吸收也有关,说明可以利用菌根真菌减少水稻砷的污染风险。Meng等[81]结合转基因技术将一种细菌的砷甲基化酶基因克隆,得到的转基因水稻的地下部和地上部均可检测到MMA(Ⅲ)和DMA(Ⅴ),其产生的挥发性砷的量比对照的野生型水稻增加了10倍,揭示可通过基因工程方法使植物具有挥发砷的能力,从而应用于砷污染土壤的生物修复与稻米的安全生产。
有研究学者认为,对于砷等潜在的有毒元素的微生物修复,在生物群落中引入新的微生物可能会引起生态失衡。所以趋向于从天然水稻土壤中分离鉴定出具有生物修复效果的微生物[82]。从水稻根际中分离鉴定出曲霉菌Pergillus sp.、青霉菌Penicillium sp.,对砷具有良好的生物挥发性,具有降低水稻籽粒中砷含量的潜力[26]。
微生物对稻田砷污染修复具有一定的应用前景,但目前投入到实际应用中比较少。土壤环境中的不稳定性,容易影响微生物的正常代谢活动。目前的微生物修复技术仍局限于实验室研究和田间实验阶段。
3 结论与展望微生物是影响土壤中砷氧化、还原和甲基化的主要驱动力,微生物对砷的氧化和甲基化作用均可以将砷(Ⅲ)氧化成毒性和生物有效性更低的砷(Ⅴ)。如何利用微生物修复砷污染土壤环境和生产安全的农产品成为国内外研究热点。微生物修复技术目前还处于田间试验和示范阶段,还需要加强对微生物影响砷转化的机理性研究和更多的田间结果来支撑该技术的研究和发展。其中,近期可以侧重下面几个方面的研究:
1) 随着基因技术的日渐成熟,已出现可以利用砷转化基因定量芯片将砷转化基因整合的技术,实现环境砷转化功能快速检测,更加全面了解环境中砷的迁移转化[83]。在日后的研究中可以利用砷转化基因定量芯片与其他组学方法和地理化学分析一起应用于砷抗性、生物转化和生物地球化学的研究中。这种多相方法可以协调地球化学和基因组学数据,以发展系统模型来预测砷的命运和影响以及环境中微生物的运动。
2) 探索微生物、植物以及土壤三者之间的相互作用机制,发展联合修复技术,关注实际应用的修复效果。只有部分微生物能够使水稻籽粒中砷含量降低到可食用的水平,说明在多数情况下,仅仅靠微生物来降低水稻籽粒砷含量达到可食用水平较难。在日后可利用接种微生物与其他修复措施相结合,如联合水稻与超(高)富集植物间套作[84]、水肥管理措施(如干湿交替、施加磷肥、硅肥)、施加类芬顿试剂[85]或其他新型修复材料修复,加强降低水稻籽粒砷效果,以便水稻籽粒达到安全可食用标准。特别是研发和应用铁基生物炭固定化微生物菌肥等复合调理剂,以及矿物基土壤纯化剂与功能微生物联用技术等,可长效固定化稳定化土壤中的砷,进而实现修复安全生产。
3) 微藻成为近几年的研究热点,可以加强研究利用微藻在砷污染稻田修复的应用,重点研究微藻对砷高效吸收与藻类回收、脱水与处置利用等技术,这将包括创新技术的发展,或对现有技术的改进。
4) 近年来,稳定同位素示踪技术迅速发展,利用稳定同位素作为示踪剂掺入到生物系统中,再利用各种手段检测它们在生物体内变化的踪迹、存留的位置或含量的应用研究发展较快。在日后的研究中可以利用稳定同位素示踪技术追踪水稻不同生育时期所累积砷的分配、转运路径和形态变化,并可进一步分析对籽粒中砷累积贡献最大的生育时期,了解水稻生长过程对砷形态转化的影响,为针对性地实施农产品安全生产的措施提供理论基础。
